Practical advice
On foot: Rome's historic centre is relatively small and can be explored on foot. You will in fact be tempted to do a great deal of walking. Pace yourself. Visits add up, and after two days you may be exhausted. Bring excellent shoes and keep the journeys as short as you can. Take the metro whenever you can (there are only two lines) and take regular breaks.
The metro: Rome has only two metro lines, and neither goes to the historic heart of the city. The only station where these two lines meet is Termini (Rome's main railway station).
The car: Forget the car in Rome. If you arrive in Rome by car, plan in advance where to park it (a hotel with parking, for example) and leave it there. Traffic is difficult, parking spaces are scarce, and you will not enjoy the city.
